| 内容記述 | We examined the effects of extremely small doses,100 mGy/day, 5 days/week, total of 12 weeks X-ray irradiation on the outer root sheath (ORS) cells of C3H/He mice (X-ray group). We also applied 1mM TEMPOL in drinking water in the same fractionation regimen (TEMPOL X-ray). And, these results were compared with control group (C) and TEMPOL control group. After the fractionated X-ray irradiation, the skin was removed and the cells in ORS were examined by H-E staining and immunohistochemistry methods. The cells with nuclear membrane abnormality and the cells with nucleus condensation change were examined. The X-ray irradiated mouse showed increased number of cells with nuclear membrane abnormality and those with nucleus condensation change in ORS. The pathological section was stained with 8-OHdG, 4-HNE, TUNEL, VEGF, HO-1, nitrotyrosine and PGP9.5 immunohistochemically. The results of immunohistochemistry showed that cells stained with 8-OHdG, 4-HNE, TUNEL, VEGF showed significantly increase in X Group. Electron microscope examination was also performed. The results showed that X-ray fractionation Group showed that the irregular shrunk image of ORS cells in X-ray group. Those results were not obtained in TEMPOL drinking group, suggesting TEMPOL scavenged reactive oxygen species initiated by X-ray fractionated irradiation. These results suggest that 100 mGy 60 times fractionation resulted in oxidative damage in ORS cells, and TEMPOL inhibited those effects. | |||||
