We also applied 1mM TEMPOL in drinking water in the same fractionation regimen (TEMPOL X-ray). And, these results were compared with control group (C) and TEMPOL control group. After the fractionated X-ray irradiation, the skin was removed and the cells in ORS were examined by H-E staining and immunohistochemistry methods. The cells with nuclear membrane abnormality and the cells with nucleus condensation change were examined. The X-ray irradiated mouse showed increased number of cells with nuclear membrane abnormality and those with nucleus condensation change in ORS. The pathological section was stained with 8-OHdG, 4-HNE, TUNEL, VEGF, HO-1, nitrotyrosine and PGP9.5 immunohistochemically. The results of immunohistochemistry showed that cells stained with 8-OHdG, 4-HNE, TUNEL, VEGF showed significantly increase in X Group. Electron microscope examination was also performed. The results showed that X-ray fractionation Group showed that the irregular shrunk image of ORS cells in X-ray group. Those results were not obtained in TEMPOL drinking group, suggesting TEMPOL scavenged reactive oxygen species initiated by X-ray fractionated irradiation.
