内容記述 In this work, we investigate the possible role of BRCA1 in telomerase
regulation in response to ionizing radiation (IR). The two human breast
cancer cell lines expressing either BRCA-null (HCC1937) or BRCA1-wild type
(MCF-7) cell lines were exposed to 4Gy of carbon ions. At different time
point following radiation, cell cycle distribution was detected by flow
cytometric analysis; telomerase activity was determined by PCR ELISA; the
expression of hTERT mRNA, a catalytic subunit of telomerase complex, was
determined by RT-PCR; the expression of telomerase protein, BRCA1 and p21
was examined by Western blotting. The results showed that a strong
down-regulation of telomerase activity, telomerase expression and hTERT mRNA
was observed in MCF-7 cells following irradiation. At the same time, IR
induced a significant G2 arrest in these cells. In contrast, neither
down-regulation of telomerase activity nor G2 block was detected in HCC1937.
Furthermore, the decline in telomerase expression was rescued when G2
checkpoint was abrogated in MCF-7. The data suggest that inhibition of
telomerase is largely BRCA1-dependent, which is mediated by G2 checkpoint.
