The two human breast\ncancer cell lines expressing either BRCA-null (HCC1937) or BRCA1-wild type\n(MCF-7) cell lines were exposed to 4Gy of carbon ions. At different time\npoint following radiation, cell cycle distribution was detected by flow\ncytometric analysis; telomerase activity was determined by PCR ELISA; the\nexpression of hTERT mRNA, a catalytic subunit of telomerase complex, was\ndetermined by RT-PCR; the expression of telomerase protein, BRCA1 and p21\nwas examined by Western blotting. The results showed that a strong\ndown-regulation of telomerase activity, telomerase expression and hTERT mRNA\nwas observed in MCF-7 cells following irradiation. At the same time, IR\ninduced a significant G2 arrest in these cells. In contrast, neither\ndown-regulation of telomerase activity nor G2 block was detected in HCC1937.\nFurthermore, the decline in telomerase expression was rescued when G2\ncheckpoint was abrogated in MCF-7.
