内容記述 | Purpose: To identify haplotypes of SNP markers associated with risk of adverse reactions after radiotherapy (RT). Patients and Methods: DNA samples were collected from 153 cervical cancer patients, who underwent conventional RT. The ninety-two patients received no chemotherapy. A total of 1201 SNPs from 173 candidate genes for radiation susceptibility were genotyped, and haplotype associations between groups were assessed. Results: Using the NCI-CTC scoring system, the patients were grouped according to adverse gastrointestinal reaction within 3 months of starting radiotherapy: a reaction grade =< 1, n = 106, and the reaction grade >= 2, n = 47. No association between any clinical factors and the adverse reactions was observed. Global haplotype association analysis (P < 0.05 and false discovery rate < 0.05) indicated that estimated haplotypes in the 8 loci were associated with risk. We revealed 4 haplotypes significantly related with adverse reaction risks. Conclusions: Individual radiosensitivity may be partly determined by these haplotypes in multiple loci. Our data could be the evidence to lead a better understanding of the mechanisms in individual radiation sensitivity. |
