内容記述 | Our previous works provided evidence that carbon-ion irradiation could repress telomerase activity via BRCA1 regulation in MCF-7 cells. It was proposed that telomerase inhibition could result in telomere shortening, followed by proliferation arrest and cell death. However, telomere shortening was not observed after X-ray or heavy-ion irradiation in our current study. To further understand the regulatory pathway between BRCA1 and telomere, DNA-PKcs, as a downstream effector of BRCA1, was investigated. Inhibition of DNA-PKcs led to significant loss of cell viability via apoptosis and senescence. It was reported that the human telomerase RNA component, hTR, stimulated the kinase activity of DNA-PKcs. These data suggested that the telomerase regulation was mediated by BRCA1 and DNA-PKcs, in-depth investigation about the effect of telomerase activity on radiosensitivity should be carried out. | |||||
