内容記述 | Accidental exposure of abdomen with high-dose radiation leads severe consequences initiated by disruption of mucosa in small intestine. The practical choice of the clinical treatment is limited, even though various methods to cure the damage have been investigated particularly in the field of regenerative therapy. To publish treatment methods that can be used for therapy readily, we examined several current pharmaceutical drugs, whose safety estimations were completed, in mice underwent severe mucosal damage by radiation. Assuming as post-accidental treatment, drugs were injected to mice 24 h after exposure at 15.7 Gy of x-ray. Effects of drugs on damaged mucosa of small intestine were evaluated by early regeneration indexes, expression of c-myb mRNA and proliferation of epithelial cells as microcolony (MC) at Days 4 and 5 after irradiation. Finally, the effects of drugs on survival rate of mice with abdominal x-irradiation at 15.7 Gy were examined. Enhancement of the mucosal regeneration at Day 4 (c-myb: P<0.01, MC: P<0.05) and improvement of the survival rate (P<0.05) were observed when clinical dose of gonadotropin, a stimulator of androgen, was injected. Similarly clinical dose of thiamazole that prevents secretion of thyroid hormone stimulated mucosal growth at Day 5 (c-myb: P<0.01, MC: P<0.05) and improved the survival rate (P<0.05). In the cases of nonclinical drugs, histamine and high dose of growth hormone antagonist, octreotide, showed significant life-saving effects (P<0.01and P<0.05, respectively). These results can be referred to construct therapeutic program and be utilized for basic studies to control regeneration of damaged mucosa. |
