内容記述 | Objective: Metabotropic glutamate receptor subtype 1 (mGluR1) regulates glutamate (Glu) excitotoxicity in CNS. Here, we aimed to monitor mGluR1 in rat with excess Glu release. Method: The rat, which intraperitoneally transplanted an Alzet osmotic pump containing N-acetyl-L-cysteine (NAC) solution, was used for this study. Extracellular Glu concentrations (EGc) were measured by microdialysis. PET assessments with [11C]ITDM, a selective radioligand for mGluR1, were conducted at several time points after the transplantation. Results: EGc in the day 1 (D1) of both groups increased compared to the pre-experiments. Following, EGc of NAC subjects kept rising until the D7. Similarly, PET study with [11C]ITDM showed retaining BPND with high level in the D7 of NAC subjects compared to the control subjects. Conclusion: [11C]ITDM-BPND is upregulated depending on increase of EGc. |
