内容記述 Density of hydroxyl radical (•OH) and hydrogen peroxide (H2O2) generated in water irradiated by carbon-ion beam was estimated under aerobic and hypoxic conditions. TBARS induced in an oil or liposome suspension samples were measured.
For •OH detection, a concentration series of DMPO water solutions (0.49‒2278 mM) were irradiated by 32 Gy carbon-ion beam. For detection of H2O2 cluster, a concentration series of TEMPOL water solutions (0.76‒1661 μM) were irradiated 0.5‒256 Gy carbonion beam according to the concentration of TEMPOL. For TBARS estimation, oil or liposome suspension samples were irradiated 256 Gy.
Intermolecular distance of sparse •OH induced by X-ray was estimated as 7.1 and 8.8 nm under aerobic and hypoxic condition, and that by 80 keV/μm carbon-ion beam was 6.3 and 7.9 nm under aerobic and hypoxic condition, respectively. Intermolecular distance of dense •OH may be different between aerobic and hypoxic condition, but details could not be evaluated from this experiment. Internal distance between H2O2 clusters was estimated as 59.0, 17.7 and 19.5 nm induced by X-ray, and 20 and 80 keV/μm carbon-ion beam under aerobic condition, respectively. However, those were estimated as 80.4, 108.9 and 97.9 nm under hypoxic condition for X-ray, and 20 and 80 keV/μm carbon-ion beam, respectively. No increase of TBARS was observed in oil or lipid membrane after 256 Gy irradiation of X-ray, and 20 and 80 keV/μm carbon-ion beam.
