内容記述 | To find the possible index for long term and low dose irradiation, the effect of repeated low dose X-ray irradiation on the melanin-derived radicals in mouse hair and tail skin was investigated. Female 8-week-old C3H/HeSlc mice were irradiated by X-ray with dose of 100 mGy/day on 5 days/week (Mon to Fri) for 12 weeks. Similarly, 4-week irradiation experiment was carried out with 500 mGy/day for C3H/HeSlc mice, or with 10, 100, and 500 mGy/day for female 8-week-old C57BL/6NCrSlc mice. An aliquot (~10 mg) of the hair sample was weighed accurately and stuffed in a plastic tube. The 2 cm tip of the tail was sampled and lyophilized. Melanin-derived radicals in hair and tail samples were measured by X-band (9.4 GHz) electron paramagnetic resonance (EPR) spectrometer. Effect of 1 mM 4-hydroxyl-2,2,6,6-tetramethylpiperidine-N-oxyl (TPL) added in drinking water on the melanin-derived radicals in hair and tail sample was tested. After the X-ray irradiation at 100 mGy/day for 12 weeks, no difference was obtained for melanin-derived radicals in the hair of the irradiated and non-irradiated groups. The X-ray irradiation at 500 mGy/day for 4 weeks makes melanin-derived radicals increased in hair compared to non-irradiated group. Melanin-derived radicals in hair may be responsible for X-ray irradiation but the threshold dose was expected; the amount of melanin-derived radicals in the tail skin was increased as dose dependently. The melanin-derived radicals in skin may be an endogenous marker for long term and low dose irradiation. | |||||
