内容記述 | Pancreatic cancer is one of the most aggressive diseases with an extremely low 5-year survival rate, and shows inherent resistance to conventional chemotherapy and radiotherapy. Carbon-ion (C-ion) irradiation has advantages with accurate dose distribution and higher tumor cell killing. To compare the effects of X-ray and C-ion irradiation on the invasive potential of MIAPaCa-2, the cells were irradiated with either X-ray (0, 2, or 4 Gy) or C-ion (0, 0.5, 1, 2, or 4 Gy), and examined the migration, invasion, MMP expression and invasiveness in response to MMP inhibitors and/or ROCK inhibitors. X-ray irradiation stimulated invasiveness and induced MMP2 expression. In contrast, C-ion irradiation suppressed invasiveness without induction of MMP-2. Treatment of cells with MMP inhibitor, GM6001, reduced X-ray-induced invasiveness, but did not reduce the basal level of invasiveness. Analysis of cellular morphology showed inter-convertible mesenchymal and amoeboid modes, which may limit the effectiveness of single inhibitor. The use of ROCK inhibitor, Y27632, in addition to GM6001, reduced both the X-ray-induced and basal invasiveness. C-ion irradiation suppressed invasiveness much more than these inhibitors. In conclusion, C-ion irradiation gives higher efficiencies compared to X-ray irradiation in diminishing the invasive potential of MIAPaCa-2 cells. | |||||
