| 内容記述 | Purpose: To investigate the medium-mediated bystander effect and its induction factor by co-cultivating unirradiated HSG neoplastic cells with HSG cells irradiated by a 290 MeV/u carbon beam. Results: An increase in cell proliferation relative to an unirradiated recipient HSG was found in the co-culture and its subculture, as well as micronucleus (MN) induction, which occurred in unirradiated recipient cells. 100 keV/mm carbon ion irradiation was found to be a much more efficient inducer of the medium-mediated bystander effect compared to 13 keV/mm carbon ion irradiation. The treatment of cells by PTIO, a specific scavenger to nitric oxide (NO), resulted in the elimination of such effects, which supports a role for NO in the medium-mediated bystander effect. As an oxidization product of NO, nitrite was indeed detected in the co-culture medium, and the dose response of its formation was also measured. Moreover, sper/NO, a NO generator, was used to directly examine the effect of NO on HSG cells. When the HSG cells were treated by sper/NO with a concentration of less than about 20 mM, cell proliferation was enhanced. However, MN was concurrently observed in the cells. Conclusions: This research first show that radiation-induced bystander effect was relative to LET; and NO, excreted from the heavy-ion irradiated HSG cells, as an important signal factor, played an dominate role in the medium-mediated bifunctional bystander effect. |
