Objectives Previous reports suggested that the tumor uptake of hypoxia PET tracers 18F-FMISO and 64Cu-ATSM was related to prognosis, but also that their intratumoral distributions were different. The purpose of this study was to compare the uptake regions of these two PET tracers with those genetically responding to hypoxia as a gold standard in mouse xenografts. Methods Human colon carcinoma HCT116 cells were stably transfected with a sodium/iodide (NIS) reporter gene controlled under 12 hypoxia responsive elements (HRE), and were injected subcutaneously into the mice. Three weeks later, dual autoradiography was performed using mixture of 64Cu-ATSM/125I– (n = 20) or 18F-FMISO/125I– (n = 12) after housing under hypoxia (10% O2, 12 hours, n = 16) or normoxia (n = 16). The uptake regions were identified objectively by cluster analyses, and the overlapped regions were quantified as percentages to 125I– uptake regions. Results The both dual autoradiographic images showed considerable areas of double tracer accumulation of the hypoxia PET tracer and 125I–. However, the overlapped regions of 18F-FMISO and 125I– uptake were significantly larger than those of 64Cu-ATSM and 125I– uptake under normoxia (54.6+-12.7% vs. 32.4+-14.7%, p = 0.008) and hypoxia (71.7+-15.3% vs. 41.9+-14.6%, p = 0.003). Conclusions The considerable overlapped regions in the both dual autoradiography supported the previous reports that the tumor uptake of 64Cu-ATSM and 18F-FMISO were both related to the resistance to therapy. However, the intratumoral distribution of 18F-FMISO closer to the genetically hypoxia responsive regions suggested that 18F-FMISO PET might be more suitable than 64Cu-ATSM PET for planning hypoxia-targeted intensity modulated radiation therapy (IMRT). |
