内容記述 | To assess the biological effects of long-term low-dose-rate gamma irradiation, the induced transcriptional changes were analyzed by DNA microarray using Affymetrix Mouse Genome 430 2.0 GeneChips. C57BL/6J mice were exposed at a dose-rate of 20 mGy/day. 1 mGy/day and 0.05 mGy/day for 400 days. The exposure levels of the dose-rates of 1 mGy/day and 0.05 mGy/day correspond to those of astronauts in space and the dose limit for radiation workers, respectively. Results showed that the expression levels of 20 genes, 11 genes and 3 genes were modulated to more than 1.5-fold in all 6 mice immediately after the irradiation at 20mGy/day, 1mGy/day and 0.05mGy/day, respectively (Radiat.Res. (2010) 174, 611-617). Next, the modulation kinetics of gene expression was investigated. The modulation of expression after irradiation at 20mGy/day was not maintained for 24 hours after the irradiation. As some effects of low-dose-rate irradiation have been reported to emerge late after the end of irradiation, the microarray analysis of the liver 3 months after the irradiation was conducted. Although there is very little overlap between the genes modulated immediately and 3 months after the irradiation, the functional analysis using Ingenuity Pathways Analysis (IPA) suggested that both gene groups were commonly related to those involved in lipid metabolism. IPA study also suggested a most significant relationship between the modulated genes and those involved in liver tumorigenesis. | |||||
