内容記述 | Fibrillary tau lesions are characteristic of diverse neurodegenerative disorders collectively referred to as tauopathies. We have been committed to development of positron emission tomography (PET) radioligands for tau fibrils in tauopathy patients and tau transgenic mice as represented by a strain termed PS19. It is noteworthy that tau deposits undetectable by PET may induce neuronal death in the PS19 transgenic line, supporting the view that oligomeric and/or protofibrillar tau assemblies are also implicated in neurotoxicity. We have recently found that vitronectin, an extracellular matrix protein, accumulates in the brain of PS19 mice, and acts protectively against tau-provoked neurodegeneration. Agents binding to vitronectin and associated molecules would accordingly offer diagnostic and therapeutic approaches to tauopathies complementary to compounds targeting tau fibrils. | |||||
