We have been committed to development of positron emission tomography (PET) radioligands for tau fibrils in tauopathy patients and tau transgenic mice as represented by a strain termed PS19. It is noteworthy that tau deposits undetectable by PET may induce neuronal death in the PS19 transgenic line, supporting the view that oligomeric and/or protofibrillar tau assemblies are also implicated in neurotoxicity. We have recently found that vitronectin, an extracellular matrix protein, accumulates in the brain of PS19 mice, and acts protectively against tau-provoked neurodegeneration.
