内容記述 | In eukaryotic cells, various organelle cooperatively works to sustain the supply chain management of cellular products and an eminent recycle mechanism known as autophagy enables this circulation of products. The system keeps homeostasis concerning energy metabolisms for cell proliferation with a proper cell cycle as well as communication among the cells constituting an organ. These concerted multi-reactions among the “intracellular creatures” look conspicuously like ecological systems that exert robustness against internal or external perturbation. Radiation stress as one of external ones would temporarily disturb the micro-world ecosystem. Although much knowledge of radiation effects on mitochondria have been accumulated, effects on and responses of other organelle including endoplasmic reticula (ER) have not yet been explored. ER contacts to mitochondrial and exchange Ca2+ ions as a signal of TCA cycle activation or apoptosis induction. Another role is folding of proteins provided by ribosomes. The properly folded proteins are taken in the transport vesicles and then delivered at Golgi body. Radiation effects on this organelle-network have been addressed in our research project, particularly focusing on ER in the hub of the network. We applied fluorescent agents to stain ER or Golgi body as well as mitochondria. Less-toxic chemicals and staining conditions were determined to perform a long-term tracking of living cells. Distribution and morphological alterations of these organelles in human cells exposed to X-rays will be presented. | |||||
