内容記述 | Radiation-induced local multiple damage to DNA (cluster DNA damage) is thought to be closely related to the biological effects of radiation. We developed a new protocol to visualize DNA damage in genomic DNA extracted from TK6 cells irradiated with a low-dose ionizing radiation by atomic force microscopy (AFM). This was realized by isolating and concentrating the DNA molecules with lesions from the whole irradiated DNA molecules also containing intact ones. We firstly applied the method to Fe ion- and X-irradiated cells. As a result, we observed a clustered DNA damage in X- and Fe irradiated genomic DNA, whereas did not found it in Fenton’s regent-treated DNA. In addition, Fe-ion beams, a high-LET radiation, produced various types of clustered DNA damage with higher complexity than those of X-rays. They also produced multiple clustered DNA damage sites in DNA. Moreover, we found that base damage or cluster damage were repaired by base excision repair (BER) mechanisms, but a flanking DSB (DSB + base damage) was repaired by homologous recombination (HR) or non homologous end joining(NHEJ). | |||||
