内容記述 | Current models of primate object vision emphasize hierarchical feed-forward connections, and recently, intracortical recurrent connections within ventral visual cortex(VVC). However, contribution of dense feedback projections from amygdala -a subcortical structure involved in emotion and social processing- has received less attention. Here we investigated role of the amygdala in socio-emotional representation in the VVC of macaque monkeys. Three monkeys received adeno-associated viral vector injections in the unilateral basolateral amygdala to express hM3Dq, an excitatory DREADD (Designer Receptor Exclusively Activated by Designer Drug). Several weeks after the injection, a positron emission tomography (PET) scan confirmed hM3Dq expression. Electrophysiological recording and FDG-PET imaging verified that administration of a DREADD-selective agonist, DCZ (1 μg/kg), significantly induced neural excitation selectively in the hM3Dq-expressing region. To examine the functional effect of amygdala excitation on the neural representation in the VVC, two of the monkeys were implanted with electrocorticogram electrode array covering the ipsilateral VVC. Visually evoked response was recorded while the monkey performed a simple fixation during a sequential presentation of stimulus images, varying in two properties: social (face or non-face) and emotional (neutral or negative). Chemogenetic activation of the amygdala attenuated the stimulus-evoked visual response to facial images in the face-selective regions in the VVC, suggesting perturbation of normal visual response in the VVC. We applied classification analysis with linear support vector machine algorithm to decode social and emotional information from the VVC visual response. The decoder successfully discriminated face from non-face and negative facial expression from neutral expression in both animals, but general emotion was discriminated only in one animal. DCZ-induced chemogenetic activation decreased the decoding accuracy of face (p< 0.001, χ2 test, uncorrected) and facial expression (p< 0.05). The similar decoding analysis was further applied on the visual response to the control stimuli varying in color and shape in one monkey. Unlike socio-emotional information, significant discrimination of color and shape information remained spared after chemogenetic amygdala excitation. These results suggest that the primate amygdala contribute to the representation of socio-emotional information in the VVC. | |||||
