内容記述 Induction of premature cellular senescence is one of mechanisms to suppress carcinogenesis, whereas senescent cells are involved in the induction of further senescence or carcinogenesis of surrounding normal cells. The evidences suggest that intracellular environment in senescent cells would be different from that of normal cells. And autophagy is well known to correlate with induction of senescence. To reveal the intracellular autophagy activities in human fibroblast cells after 20Gy X-ray exposure and to understand the relationship between the senescence and autophagy, we analyzed the expression level of the four proteins, p16 and p21, which are involved in senescence, and LC3 and pp62, which play a role in autophagy, by western blotting of extract from irradiated cells.
The expression level of p16 and p21 gradually increased after irradiation. Although the expression level of LC3 also increased until a few days after exposure, pp62 decreased rapidly. It is inferred that pp62 is necessary for selective autophagy, but be decomposed after the autophagy. These results indicated that selective autophagy could be activated immediately after irradiation, and then, non-selective autophagy could be activated and cause secretion of certain factors inducing late effects, such as carcinogenesis of surrounding cells. The processes might regulate intracellular homeostasis. For further study, application of a novel technique using nano-diamonds will be discussed in terms of sensing physical parameters, such as temperature, in the senescent cells. Preliminary measurement of temperature was performed using EGF-binding NV-nano-diamonds will be shown.
