内容記述 The goal of this project is to evaluate the role of deficiencies in DNA repair pathways on particle therapy (PT) response. We have exposed five cell lines (M059J, M059K, H1299, H460 and BxPC-3) to 6 MV x-rays, protons (100 MeV: 1.1, 2.5 and 7.8 keV/µm), He-ions (150 MeV/u: 2.2, 6.9 and 13.4 keV/µm) and C-ions (290 MeV/u: 13.5, 28.1 and 61.4 keV/µm) for clonogenic cell survival assessment. The response of the cell lines were then characterized in terms of the relative biological effectiveness (RBE). Here we presented updated values of the RBE of these cell lines, which ranged from 0.90-1.70 for protons, 0.87-1.30 for He-ions, and 1.11-1.97 for C-ions. Our data on RBE indicated that different cell lines respond widely differently to PT beams. We further demonstrated that DNA repair deficiencies play a major role in the response of cell lines to PT beams. Particularly, our data suggest that non-homologous end joining repair has a major role in repairing radiation induced DNA lesions.
