内容記述 Cellular senescence is a state of persistent proliferative arrest and may prevent the cells from entering tumor processes. Senescent cells secrete growth factors and inflammatory cytokines. Maintaining chronic inflammation may promote pro-tumorigenic properties, thus senescent cells might effect on formation of tumor microenvironment. In radiotherapy, secondary primary cancer is potentially induced by unexpected exposure to normal fibroblasts. It would be important to reveal the correlation between the senescent fibroblasts and their role in cancer induction.
The exposure to ionizing radiation causes damage to not only DNA but also proteins or organelles in cytoplasm of cells. Accumulation of damaged proteins and dysfunctional organelles are thought to lead to cell death, such as apoptosis. However, irradiated fibroblasts are highly resistant to apoptosis and they maintain their viability entering senescent state. This suggests that, in irradiated fibroblasts, autophagy eliminates accumulated the damaged proteins and the dysfunctional organelles to avoid induction of apoptosis. In this study, to clarify the role of autophagy on cell survival, we investigated molecular processes of autophagy and senescence in irradiated fibroblast cells.
Normal human fibroblasts BJ-5ta cells immediately arrested cell proliferation and induced cellular senescence after exposure to X-rays. Expression of LC3-II transiently increased after irradiation and then gradually decreased to the control level. It gradually rose again with induction of senescence. The expression of phosphorylated ULK1 was correlated with that of LC3-II. These results suggest that early induction of autophagy after irradiation have different roles from re-activation of autophagy with induction of senescence.
