内容記述 | DREADDs (Designer Receptors Exclusively Activated by Designer Drugs) are unique neuromodulatory tools which can inhibit or activate the hosted neurons via systemic administration of designer drug (clozapine-N-oxide, CNO). Without any implants or probes, DREADDs allow us to examine the relationship between target neural system and freely moving animal behavior. Here, we aimed to manipulate the nigrostriatal dopamine system in common marmosets (Callithrix jacchus) monitored by multimodal positron emission tomography (PET) imaging. One common marmoset received an injection of AAV-CMV-hM3Dq vector (modified form of the human M3 muscarinic receptor) into the left substantia nigra (SN) and another control vector injection into the right SN. At fifty-six days after the injection, PET imaging with a DREADD-selective ligand, [11C]X visualized hM3Dq expression in ipsilateral SN and striatum. Be indicated that CNO intravenous pretreatment increased neural activity in SN. PET imaging with [11C]raclopride, a selective dopamine D2 receptor antagonist, also suggested that dopamine release was upregulated by intravenously pretreatment of CNO in the ipsilateral striatum. These results demonstrated that chemogenetic techniques combined with PET imaging are applicable for manipulation of nigrostriatal dopamine system in common marmosets, which help us to understand its role for social communications or interactions. | |||||
