内容記述 | Protection of normal tissues adjacent to the tumor from radiation injury is of critical clinical importance in cancer patients undergoing radiotherapy. However, the clinical application of radioprotectors, such as amifostine, is limited due to their toxicity. We have synthesized a planar catechin, where an electron-donating isopropyl group is introduced into the catechol moiety natural occurring (+)-catechin to improve its radical-scavenging activity. In this study, we conducted some in vitro and in vivo experiments using rat primary thymocytes, normal human fibroblasts, and mice to evaluate biological effects of the planar catechin. In addition, the radical-scavenging activity of the planar catechin was estimated by fluorescence method and electron paramagnetic resonance (EPR) spin trapping method. The planar catechin efficiently protected the thymocytes and the normal human fibroblasts against radiation-induced cell death. The planar catechin has a high scavenging ability for hydroxyl radicals induced by radiation and inhibits radiation-induced DNA cleavage. We also assessed the oral use of the planar catechin in mice before X-ray treatment. Our results demonstrate the possibility of using the planar catechin as an effective radioprotector in radiotherapy. | |||||
