内容記述 (Objectives) Sake is a popular Japanese alcoholic beverage, recently spreading to the world. Moreover while drinking custom of sake a lot has negative effects on our health, the beneficial effects on health also attract attention. It remains unknown whether or not chronic sake drinking custom is related to other risk factors such as radiation. Effects of chronic intake of alcoholic beverages on radiation-induced metabolic alteration were evaluated using Japanese sake.
(Materials & Methods) As Japanese sake, junmai-shu, (ethanol content: 15%) was used. Sake was administered to female mice(C3H/He) by using a feeding needle every morning for four weeks (8.7 ml/kg or 26 ml/kg), and fractionated irradiations were performed with 0.75Gy/day for the last four days of the intake period. Mice were euthanized one day after the last irradiation and the liver and blood were removed. As a comparative study, 15% ethanol water was administered to mice instead of sake. Liver samples was extracted and analyzed by the cation mode of CE-TOFMS (Agilent Technologies). Primary Component Analysis was conducted about the result and influential candidate molecules were selected and evaluated. Metabolic markers in the serum were also analyzed using DriChem7000V (Fuji Film).
(Results & findings) As a result of primary component analysis of MS analysis in liver, radiation accompanied by chronic intake of sake caused remarkable changes in anti-oxidative functions and energy metabolisms, which were not altered by radiation or the sake intake alone respectively. Interestingly, the changes in anti-oxidative functions were not observed in the treatment of radiation accompanied by chronic intake of 15% ethanol water. Moreover, changes in some metabolites by radiation alone disappeared by chronic intake of sake. Concerning metabolic markers, though chronic intake of sake increased the amounts in triglycerides, radiation suppressed the increase.
(Conclusion) It was suggested that moderate drinking sake promotes anti-oxidative functions after exposure to radiation, possibly resulting in preventing adverse effects.
