内容記述 | 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical is a relatively stable radical and frequently used as a model of reactive oxygen species to evaluate the radical-scafenging activity of antioxidants. However, the poor solubility of DPPH radical to water has precluded its use in aqueous systems. In this study, we used beta-cyclodextrin (CD) to solubilize DPPH radical in water. Ball milling of the mixture containing DPPH radical and beta-CD for 1 h, followed by an addition of water and filtration with a membrane filter gave a purple solution. This solution showed an absorption band at 525 nm, which is diagnostic of DPPH radical. When ascorbic acid was added to the solution, the band at 525 nm disappeared immediately. This indicates that ascorbic acid efficiently scavenge DPPH radical in water. Thus, DPPH radical solubilized by beta-CD in water may be a powerful tool to evaluate antioxidative activity of antioxidants in aqueous systems. |
