内容記述 We constructed the new treatment facility for the advanced carbon-ion therapy applying a 3D fast spot scanning system with pencil beams at HIMAC in NIRS. To identify biological effects of the system, we demonstrated the cell killing of different human tumor cell lines using the spot scanning of carbon-ion microbeams generated with Takasaki Ion Accelerators for Advanced Radiation Application in JAEA and irradiated just 0.04% of total cells by the highly controlled microbeam. The percent survival in the cells with wild-type P53 gene was 89%, while almost 100% was observed in the cells with mutated-type P53 gene. We can expect the percent of 99.96% when the cell killing was induced by the just irradiated 0.04% cells. However, the 89% was significantly low beyond our estimation. Moreover, it returned to 100% when using a specific inhibitor of gap-junction mediated cell-cell communication. There is clear evidence that the spot scanning irradiation of carbon ions enables the enhanced cell killing in wild-type-P53 cells via gap-junction mediated bystander effect. (This is the co-operative study with Drs. T.Funayama, Y.Mutou, M.Suzuki, H.Ikeda, Y.Hattori and Y.Kobayashi in JAEA.)
