内容記述 To investigate differences in chromosome aberrations for G0/G1 and G2/M phase cells exposed to X rays and heavy ions with various LETs using fluorescence in situ hybridization method.
Confluent normal human fibroblast AGO1522 cells were irradiated with X rays, 490MeV/nucleon silicon (55keV/um), 500MeV/nucleon (200keV/um) and 200MeV/nucleon (440keV/um) iron ion beams, then incubated at 370C for 24hours for repair. The interphase chromosome analyses were performed by chemically-induced premature chromosome condensation (PCC) which allows scoring of aberrations in G2 phase cells, and fusion-induced PCC with mitotic HeLa cells by sendai virus for G0/G1 phase cells. Chromosome aberrations were scored by fluorescence in situ hybridization using chromosome 1 and chromosome 3 probes. Cell cycle distribution was also studied by flow cytometry at different incubation times following subculture after 24hr repair incubation in irradiated G0 cells.
Numbers of chromosome aberrations and fragments in G0/G1 phase cells are higher than those in G2/M phase cells either irradiated with 2 Gy of X-rays or heavy ions with different LETs; the fragment frequency for 2Gy X ray was very low. The color junctions observed in G0/G1 phase cells irradiated with X-rays, 55keV/um silicon ions and 440keV/um iron ions are also higher than those in G2/M phase cells. In contrast, there is no difference in color junction frequency for samples exposed to 200keV/um iron ions between G0/G1 phase and G2/M phases. The chromosome aberration, fragment and color junction frequency is LET dependent up to 200 keV/um, and then decreased for 440keV/um iron ion beams either in G0/G1 or G2/M phase. The percentages of cells in G0/G1 phase irradiated by iron ions (200keV/um and 440keV/um) after 24 repair incubation then subcultured are more than those irradiated by X rays or silicon ions.
Chromosome aberrations in G2/M phase may be underestimated in normal human fibroblasts irradiated by low or high LET heavy ions due to a high rate of G1-arrest.
