内容記述 Malignant pleural mesothelioma(MPM) is an aggressive tumor arising from serous surfaces and often related to asbestos exposure. MPM is resistant to various forms of therapy, such as radiotherapy, surgery or chemotherapy . So far, no effective therapeutics including chemotherapy or radiotherapy has been established for the disease-advanced case. The present study was carried out in order to examine the radiosensitivity and mechanism of lethal effect of MPM cell lines by X rays and carbon-ion beams. Officially distributed 6 kinds of MPM cell lines were used and irradiated with X-ray or carbon-ion beams (13.3keV/µm and approximately 80keV/um) at the Heavy Ion Medical Accelerator in Chiba. Cell-killing effect was detected using a colony-formation assay and apoptosis was evaluated by fluorescent staining with Hoechst33342. Based on the survival curves, the D10 values (one of parameters for cellular radiosensitivity) were calculated and ranged from 3.01Gy to 5.74Gy for X-rays;2.16Gy to 4.18Gy for 13keV/um carbon-ion-beam irradiation and 0.97Gy to 2.36Gy for 80keV/um carbon-beam on each cell lines. The relative biological effectiveness (RBE) values calculated by the D10, relative to X-ray, range from 1.25 to 1.62 for 13keV/um;m-ion-beam and from 2.42 to 3.10 for approximate 80keV/um-ion- beam irradiation on each cell line. Those results indicated that high LET-carbon-ion irradiations are also significantly more effective in cell killing for the 6 kinds of MPN cell lines. But the incidence of apoptosis is the very low even when cells were irradiated with 80keV/um-carbon-ion beams. So the apoptosis was not the mechanism of lethal effect by heavy ions.
