内容記述 | <Objective> In vivo imaging of genetic response to hypoxia utilizing hypoxia response elements (HRE) has been limited to the applications of optical or viral thymidine kinase reporter genes. In this study we performed it using the human NIS reporter gene in mice. \n<Method> Human colon carcinoma HCT116 cells were stably transfected with NIS reporter genes driven by a minimal promoter plus 12 copies of HRE, and were injected subcutaneously into 8 nude mice. When the tumor diameter reached 5 mm about 2 weeks later, SPECT imaging of the tumors was performed for 1 hour after intravenous injection of 99mTcO4- using a pinhole SPECT/CT system, following 12-hour housing of the mice under a hypoxic (10% O2; n=4) or normoxic (21% O2; n=4) condition. \n<Result> All mice exposed to hypoxia showed high tumor uptake of 99mTcO4-, while those kept in normoxia did not. A semiquantitative analysis of the total 99mTcO4- activity into the tumors demonstrated a statistically significant difference between the 2 groups (0.45+-0.2 vs. 0.79+-0.15, p = 0.039). \n<Conclusion> We successfully demonstrated the feasibility of molecular genetic imaging of hypoxia response using NIS reporter gene in a mouse subcutaneous xenograft model. Since NIS reporter gene has advantages over optical and previous PET reporter genes, the current animal model may be useful for investigating genetic response to hypoxia in vivo. |
