内容記述 | For biological dosimetry by counting chromosome aberrations, automation technique has been required to process large number of sample preparations at low dose radiation exposure. The metaphase finder is an automated optical microscope system, which automatically scans and finds metaphase cells on the slide glass, and relocates metaphase cells to the center of the field of view of the microscope to observe chromosomes in high magnification We have demonstrated a cost-effective metaphase finder system by assembling commercially available components, such as microscope, motorized sample stage, personal computer and general purpose image analysis software package, instead of special dedicated system, and the software has utilized the mathematical morphology based image processing to the algorithm to find metaphase cells, in last time of this meeting (1). But, conventional cell preparation has two problems: It is not capable to high dose irradiation, and the long time to observation is required. Then, premature chromosome condensation (PCC) is used instead. PCC is the phenomenon that an interphase cell to produce condensed chromosomes prematurely, with specific inhibitors of serine/threonine protein phosphatase, such as Okadaic acid and calyculin A. We had applied the induction of PCC in human peripheral lymphocytes and developed a biodosimetry on the basis of scoring ring chromosomes (2). We have used this method in 3 severely exposed patients in the Tokai-mura criticality accident. This year, we evaluate the performance of the metaphase finder on the cells that made by PCC. |
