Pancreatic cancer is a highly metastatic tumor with poor response to conventional radiotherapy. C-ion irradiation has advantages compared to conventional photon therapy with accurate dose distribution and higher tumor cell killing, and is expected to improve the clinical outcome for pancreatic cancer patients. The aim of this study was to compare the biological effects of carbon-ion (C-ion) and X-ray irradiation of the invasive pancreatic cancer cell line, MIAPaCa-2. We demonstrated that C-ion irradiation suppresses invasiveness without induction of active MMP-2 compared to conventional X-ray irradiation, which stimulates invasiveness and induces expression of active MMP-2 with enhancing MMP2 transcription. Treatment of cells with an MMP inhibitor diminished X-ray-induced invasiveness but not basal level of invasiveness. We demonstrated that the involvement of inter-convertible mesenchymal and amoeboid modes of motility in the invasiveness of MIAPaCa-2 cells limits the effect of invasiveness of single inhibitors. The use of ROCK inhibitor, in addition to MMP inhibitor, reduced both X-ray-induced and basal level of invasiveness. C-ion irradiation reduced invasiveness much more than these inhibitors and blocked the basal invasiveness of MIAPaCa-2 cells without the use of inhibitors. C-ion irradiation gives higher efficiencies compared to X-ray irradiation in diminishing the invasive potential of MIAPaCa-2 cells.
