内容記述 Radiation-induced hair loss is a clinically important, but under-researched topic. We previously created an FGF1:FGF2 chimera (FGFC) that showed greater structural stability than FGF1. FGFC was capable of stimulating epithelial cell proliferation much more strongly than FGF1 or FGF2 even in the absence of heparin. Therefore, FGFC was expected to have greater biological activity against cutaneous damage. The study aimed at developing an in vivo assay system for radiation-induced hair follicle dystrophy, and using it to evaluate the protective activity of FGFC and FGF1. A portion of the dorsal skin of 8-week-old male BALB/c mice that is in their resting phase was depilated to induce synchronized growth of the hair follicles. At 6 days after depilation, mice received total body irradiation (TBI) with gamma-rays at doses in the range from 4 to 12 Gy. Pathological changes were detected progressively in the putative growing hair follicles over the time course after TBI and the dystrophy was evaluated on the basis of stage-specific parameters reported previously, which were found to be well-suited for classification of the radiation-induced hair follicle dystrophy. In this system, it was found that FGF1 effectively prevented hair follicle apoptosis in mice in the presence of heparin. In addition, FGFC was more effective than FGF1 in preventing hair follicle apoptosis in the absence of heparin; however, radioprotective effectiveness of FGF1 was drastically diminished without heparin. These findings indicate that FGFC exerts radioprotective activity on hair follicles independently of heparin. The results further suggest that FGFC may be useful for the prevention and treatment of radiation-induced hair loss.
