内容記述 | Melanocytes characterized by the activities of tyrosinase, tyrosinase-related protein (TRP)-1 and TRP-2 as well as by melanosomes and dendrites are located mainly in the epidermis, dermis and hair bulb of the human skin. However, the mechanism of the proliferation and differentiation of human epidermal melanocytes is largely unknown. It is investigated in this study what factors are involved in regulating the proliferation and differentiation of human epidermal melanocytes derived from the primary culture of foreskin using serum-free culture. Dibutyryl adenosine 3':5-cyclic monophosphate (DBcAMP) and basic fibroblast growth factor (bFGF) induced the dedifferentiation of melanocytes in serial cultures. Melanocytes gradually lost their pigments and became melanoblast-like cells in this medium. Of growth factors and cytokines tested, endothelin-1 (ET-1) and stem cell factor (SCF) stimulated the proliferation of these melanoblast-like cells in this medium. ET-1 stimulated the differentiation of human melanocytes in a medium supplemented with DBcAMP and/or L-tyrosine. Moreover, ET-1 and SCF synergistically stimulated the proliferation of human epidermal melanocytes in this medium. These results suggest that ET-1 and SCF are involved in regulating the proliferation and differentiation of human epidermal melanocytes in cooperation with cAMP, bFGF and L-tyrosine. | |||||
