内容記述 Ecosystem functions are controlled by a variety of biotic and abiotic interactions in combinations. Changes in those interactions which caused by exposure to environmental stimuli can affect ecosystem functions. Those interactions can also influence gene expressions of individual organisms in the environment. Therefore, it is important to understand the whole gene expressions of community members for the ecological impact assessment of environmental stimuli.
We applied a High Coverage Expression Profiling (HiCEP) method to study radiation effects on eukaryotic microbial community. This method, which was developed in the National Institute of Radiological Sciences, is a transcriptome fingerprinting technique based on an Amplified Fragment Length Polymorphism (AFLP). The HiCEP method can detect a slight expression change of transcript-derived fragments with high coverage by refinements of the selective PCR technique.
Microbial community from a flooded paddy soil was exposed to gamma radiation (6 Gy), and poly(A) RNAs extracted from the community was subjected to the HiCEP analysis. We found 14,169 HiCEP peaks for the irradiated community. Among these peaks, 250 peaks were upregulated by the gamma irradiation and 85 peaks were downregulated. Up or down regulated peaks (3 and 4 peaks, respectively) were isolated and sequenced. The upregulated transcripts had a significant sequence similarity with ubiquinol-cytochrome C reductase, and the downregulated transcripts included leucine dehydrogenase and dipicolinate synthase. The results suggest that the HiCEP method is a useful tool for the transcriptome analysis of eukaryotic microbial community in the environment.
