内容記述 Although the public todeay could be exposed to X-rays as high as 1 cGy due to diagnostic procedures, the biological effects of this low dose range have not been established.In order to detect any molecular response to the corresponding level of ionizing radiation, we searched more than 23,000 transcripts in normal human fibroblassts, HFLIII. HiCEP technique revealed that more than two hundred genes were upregulated transiently be 1 cGy of X-rays during the one hour period after irradiation. The transcripts upregulated with the greatest rate of increase in the irradiated HFLIII cells encoded CXC chemokines (CXCL1, CXCL2, CXCL6). HFLIII cells were irradiated with X-rays at 10-100 mGy (60 mGy/min) and the culture media collected at 1, 2, 4, 8 hr were examined for CXCL2 (Gro-beta) protein by immuno-assays. CXCL2 ligand was increased at 2-3 folds within 4 hours after X-ray irradiation at 3-5 cGy although the same effect was not marked at 10 cGy. The data confirmed our previous results that HFLIII cells upregulate a distinct subset of CXC chemokines to the extremely low level of X-rays. We further investigate some possible signaling mechanisms iinvolved in the cellular response.
