内容記述 | Although the public today could be exposed to X-rays as high as 1 cGy due to diagnostic procedures, the biological effects of this low dose range have not been established. We searched through more than 23,000 transcripts in normal human fibroblasts, HFLIII, using a novel comprehensive expression analysis method, HiCEP. More than two hundred genes were upregulated transiently by 1 cGy of X-rays during the one hour period after irradiation. The transcripts upregulated with the greatest rate of increase in the irradiated HFLIII cells encoded CXC chemokines (CXCL 1, CXCL2 and CXCL6). In order to test the involvement of CXC chemokines in cells irradiated with low doses, we irradiated HFL III cells with 1-5 cGy X-rays, and transferred the media from HFL III culture to two melanoma cell lines characteristic of excessive numbers of the CXC chemokine-specific receptors. The growth of these melanoma lines were significantly stimulated after the addition of irradiated HFL III medium. Serum-starved HFLIII cells were irradiated by X-rays at 1-10 cGy. The culture media was collected at 1, 2, 4, 8, hour and the amount of CXCL2 (Gro-beta) protein was measured by ELISA assays. Consistently to our previous by-stander experiments using melanoma cell lines, CXCL2 was increased at 2-3 folds in 4 hours after X-ray irradiation at 3-5 cGy although the same result was not marked at 1 and 10 cGy. Our results indicate that HFLIII cells respond to doses of radiation as low as 5 cGy in a dose-independent manner. Mechanisms alternative to those involved in moderate/high dose studies have to be considered in understanding the biological effects of such diagnostic level radiation. | |||||
