cells and melanocytes by melanocyte stimulating hormone
\nAlpha-melanocyte stimulating hormone (MSH) is important humoral factor involed in regulating the melanogenesis of mammalian melanoma cells and melanocytes. The present study was designed to clarify the mechanism of the regulation of melanogenesis of mouse melanoma cells and melanocytes by alpha-MSH. Melanin synthesis and tyrosinase activity of cultured mouse melanoma cells and neonatal melanocytes were measured. The expression of the melanocyte-specific alpha-MSH receptor gene, melanocortin receptor-1 (MC1R) as well as the proopiomelanocortin (POMC) gene was analyzed by the reverse transcription-polymerase chain reaction (RT-PCR). Alpha-MSH was shown to stimulate the melanogenesis of B16-G4 mouse melanoma cells. Similarly, alpha-MSH added to serum-free primary culture of melanoblasts derived from epidermal cell suspensions of 0.5-day-old C57BL/10JHir mice induced their differentiation. The expression of the MC1R gene had already been initiated before addition of alpha-MSH, and, in addition, no up-regulation of the MC1R gene was observed after addition of alpha-MSH. However, no expression of the POMC gene was observed before or after the addition of alpha-MSH. The expression of the MC1R gene was first observed in the epidermis of 13-day-old embryos, and gradually increased up to 0.5 days after birth, and thereafter remained constant. However, no expression of the POMC gene was observed at any age of mice tested. Alpha-MSH is important factor for the regulation of melanogenesis in melanoma cells as well as the induction of the differentiation of neonatal melanocytes.
