内容記述 We have been studying bystander lethal and mutagenic effects in normal human fibroblasts irradiated with low-density carbon-ion beams generated by Heavy Ion Medical Accelerator in Chiba (HIMAC) at National Institute Radiological Science (NIRS) in Japan. Normal human skin fibroblasts were cultured on Mylar film (2.5micrometer in thickness) of 6.5mm in diameter drilled in plastic tissue culture dishes (35mm in diameter) and irradiated with 6MeV/n carbon-ion beams at the fluence density of 1.5 x 105 particles/cm2 as follows:
(1) Irradiate with all cells on Mylar film
(2) Irradiate with half of cells on Mylar film
(3) Mix with all irradiated cells and un-irradiated control cells (1:1)
(4) Treated with the inhibitor of cell-cell communication to half irradiated cells
The results of both cell killing and mutation induction at HPRT locus showed that there observed in the same effects between all-cell irradiated (1) and half-cell irradiated (2) group. Furthermore, both biological effects for half-cell irradiated group treated with the inhibitor of cell-cell communication (4) showed the same level for mixed population with all irradiated cells and un-irradiated control cells (3). There is evidence that bystander effects of both cell killing and mutation induction occurred in un-irradiated half bystander cells of the irradiation procedure (2) and cell-cell communication may play an important role of inducing the bystander effects.
