内容記述 | Aim: Nitric oxide (NO) is an essential biological molecule with a wide variety of functions in physiological and pathophysiological events. We demonstrated previously that administration of NO scavenger or NO synthase inhibitor decreased the incidence of mammary tumors of rat irradiated with gamma-ray. Thus NO might participate in the radiation-induced tumorigenesis. To elucidate the role of NO in cellular and molecular events, we examined the effect of NO on the expression and localization of tight junction-associated proteins in a mouse mammary epithelial cell line (HC11). Methods: Monolayer of confluent HC11 was incubated with either NOC5 (20 mM) or NOC18 (0.4 mM), NO donors, for 90 min or 48 hr, respectively, and used for analyses of cell-cell junctional proteins by immunochemical and immunocytochemical means. Results and Discussion: Continuous localization around the cell perimeter of occludin and ZO-1, components of tight junction, was disorganized, and descending expression of these tight-junctional proteins was revealed in HC11 treated with NOC18. On the other hand, tyrosine phosphorylation of several proteins was induced in a non-ionic detergent insoluble fraction of the cells incubated with NOC5. Furthermore, immunocytochemical observation for the phosphorylated proteins in the NOC5 treated cells showed distinct labeling at cell-cell junction sites. These results suggest that NO influences upon expression and localization of tight junctional proteins in mammary epithelial cells, and that the phosphorylation of these proteins may participate in functional modification of tight junction. |
