内容記述 Metaphase Finder is the automated optical microscope system, which automatically scans and finds metaphase cells on the slide glass, and relocates metaphase cells to the center of the field of view of the microscope to observe chromosomes.
For biological dosimetry by counting dicentric chromosomes, which requires large number of sample preparations at low dose, we have developed an automated chromosome analysis system in 1993. This system was able not only to find metaphase cells, but also to acquire enlarged image of individual metaphase cells and detect dicentric chromosomes. The analysis result achieved with assistance by this system showed very good accordance with the results by human observers alone. However, the system was too expensive, too complicated and the speed of analysis was not enough for the practical use.
Now, we have constructed an improved system, which has almost the same function of the previous prototype system, but faster, smaller and less expensive. The components for this new system, such as microscope, automated stage and PC, are commercially available products. The system has high cost-effectiveness and the flexibility in adapting to the new staining methods.
This Metaphase Finder system consists of following components: an optical microscope (Nikon Eclipse E800, equipped with 10x and 100x objective lenses), a motorized sample stage (Sankei Corp., driven by X and Y axis stepping motors of 1microm resolution), an auto-focusing unit (Flovel AF/sigma, a system of video signal processor and focusing motor) and two CCD cameras for focusing (Nikon HV-D28S, high-sensitivity 3-CCD color NTSC video camera) and for image acquisition (Hamamatsu C4742-95, 1024x1024 pixel cooled CCD camera). These components are controlled by the personal computer (Windows 98 PC with Intel Pentium III 800MHz processor).
The image recognition software for detecting metaphase cells from the microscope image was developed on the programming environment provided by the general purpose image analysis software (Image-Pro Plus, Media Cybernetics), using Microsoft Visual BASIC language.
The processing speed of this Metaphase Finder system is 20 to 30 minutes per one slide glass (typical scan area is 20x50mm^2^), which is remarkably improved from several hours per slide in the case of previous model. The algorithm for detecting metaphase cells has also improved to be less sensitive to the condition of the sample preparation.
This new system is scheduled to be distributed to several laboratories in Japan, and will be tested for the practical use.
