内容記述 | For complement to uncertainties in the epidemiological approaches to the risk estimation on human cancers due to intake of plutonium compounds, animal studies are essential to clarify variable factors related to biokinetics, dosimetry, and biological responsiveness. In the present paper, the effectiveness for pulmonary carcinogenesis of inhaled plutonium and the specific pathogenesis of the injected plutonium are summarized. (1) Pulmonary carcinogenesis in rats following inhalation exposure to plutonium dioxide: Female WM/Nrs rats exposed to high-fired (1150 degrees Celsius) 239PuO2 aerosols with AMAD of 0.3-0.4 um. Initial lung deposition (ILD) was determined by whole-body counting of LX-ray (17 keV) on day 7 after the exposure. Cumulative alpha dose was calculated by the time integral of ILD and the retention function. The incidence of lung carcinomas significantly increased over 0.45 Gy, reaching the maximum of 90% at 6.6 Gy. Histological types of the primary lung tumors were mostly epithelial types derived from bronchiolar or alveolar lining epithelium, classified into adenomas, adenocarcinomas, adenosquamous carcinomas, and squamous cell carcinomas. The differential dose responses were clearly noted among these tumor types. (2)Carcinogenesis in mice following injection of plutonium citrate: Female C3H/HeN, C57BL/6J and B6C3F1 strains of mice were intraperitoneally administered with 100 to 10,000 Bq/animal of monomeric 239Pu citrate solution. The cumulative skeletal dose until death of each animal calculated by the time integral of injected activities and the retention function. Bone tumors increased in a dose-dependent manner to a maximum incidence of 50-63% at 2-3 Gy, and most of the tumors from all the strains of mice were osteosarcomas. Lymphoid neoplasms appeared early in all the strains of mice received 5000 Bq or more and were characterized by pre-B-cell lymphomas derived from progenitor B-cells. The other solid tumors rather decreased in all the strains of mice received 500 Bq or more. |
