内容記述 NIRS (National Institute of Radiological Sciences, Chiba, Japan) owns HIMIC (Heavy Ion Medical Accelerator in Chiba) which has an accelerator to generate various heavy ion beams, and has treated over 3000 cancer patients with carbon ions. It is well known that high LET (50-200keV/um) heavy ion irradiation has strong relative biological effectiveness (RBE), and low LET (for example, less than 20 keV/um) heavy ion irradiation may not be different from low LET photon irradiation such as X-rays or gamma-rays. In order to better understand the repair system induced by heavy ion irradiation, we tested radiosensitivities of various DNA repair deficient CHO mutant cell lines (WT, DNA-PKcs, Ku80, XRCC1, XRCC2, Rad51D, FANCG) exposed to x-ray, low and high LET carbon ion irradiation by colony formation assay. Asynchronous cells were irradiated by low LET carbon ion (LET: 13keV/um), high LET carbon ion (LET: 70keV/um), and x-rays. All except DNA-PKcs and Ku80 mutants showed similar pattern of cell killing. High LET irradiation induced the highest levels of cell killing, and the low LET carbon ions provided the similar levels of cell killing to X-ray. Interestingly, Ku80 mutant cell line xrs-5 showed no LET effects in their radiosensitivities. To our surprise, DNA-PKcs mutants, V3 and irs20 which are sensitive to x-rays, showed a higher sensitivity to both low and high LET carbon ions than x-rays. We also tested a scid cell line and observed the similar effect. These results indicate that the low LET heavy ions (13 KeV/um) produce different types of DNA damages compared to X-ray. It appears that DNA-PKcs has a special function in repairing intermediately complex damage which could be caused by low LET heavy ion irradiation. The nature of these damages remains to be elucidated.
