内容記述 Biological effect of toxic reagents at low dose is not well characterized. Reliable quantitative and mechanistic information is necessary to assess the risk of the pollutants at low doses. Recent advances have enabled us to use a comprehensive approach in biological studies of humans and other species. We developed a novel gene expression profiling method called high coverage expression profiling, or HiCEP. By applying the new technology, we investigated the effects of low-dose ionizing radiation (0.01 Gy) and sodium arsenite (1 microM) on primary normal human fibroblasts. Transcripts most upregulated by the low dose X-ray were revealed to be C-X-C chemokines, which mediate extracellular signaling and are involved in certain type of malignancy (Fujimori et al., Cancer Research 65: 10159-10163, 2005.). Transcripts upregulated by sodium arsenite at the regulation level includes HMOX-1 and other known and unknown mRNAs. The strategy successfully detected the very small alteration of the gene expression, thus promising as a useful tool to develop risk assessment for very low levels of environmental toxic reagents. Another advantage of HiCEP is that it can be applied to most animals and plants on the earth regardless of the availability of their genome information, which will promote us to explore many useful biomarkers.
