内容記述 | The F1 mice between gpt delta transgenic and SWR strains were exposed to gamma ray at three kinds of dose rate (average 0.04255 mGy/day, 0.858 mGy/day and 16.56 mGy/day) for 483 days. Total doses received by those mice were approximately 20, 400 and 8000 mGy. Chromosome preparations were made from spleens of 6, 7, 8 and 6 mice irradiated at those doses of 0 (control), 20, 400 and 8000 mGy, respectively The single cell suspensions obtained from those spleens were cultured for 43 - 46 hrs in the RPMI1640 medium containing fetal calf serum (20%), Concanavarin A (3 micro g/ml) and Lipopolysaccharides (10 micro g/ml). Colcemid (0.03 micro g/ml) was also added to the culture medium for last 24 hours before the harvest. Quinacrine / Hoechst 33258 double staining method to detect both the chromosome banding pattern and centromere position was applied to the analysis of dicentrics and fragments. As all of mouse chromosomes are acrocentrics, considerable number of fused or associated chromosomes similar to the dicentrics was observed. Therefore, only typical dicentrics accompanied by a fragment was scored in the present study. The total number of cells analyzed in the mice irradiated at the doses of 0, 20, 400 and 8000 mGy were 1254, 1537, 1659 and 1259, respectively. The frequencies of the typical dicentric chromosomes per metaphase in those cases were 0, 0.0019, 0.0018 and 0.02, respectively. It may be worth to be mentioned that radiation effect was detected in the mice received 20 mGy by the irradiation at the dose rate of 0.04255 mGy per day. Now we are analyzing more cells to be able to show significant dose effect in those mice. |
