内容記述 We have investigated biological responses in vitro and in vivo of heavy ions including carbon ions. Biological questions involved in clinical carbon ion therapy consist of two items; namely, (1) how biological gain of carbon ions could be increased, and (2) whether biological effectiveness of carbon ions depends on tumor types. The first question arose from the anxiety that high LET radiotherapy would cause severe normal tissue damages and annihilate therapeutic gain. As RBE values of different cells in vitro are not unity, which has been reported by extensive studies conducted by Dr. Ellie Blakely, comparison of RBE between tumor response and normal tissue reaction would clarify biological gain of carbon ions. We decided to investigate direct comparison of RBE values between tumor growth delay and skin reaction. The second question comes from the initial observation of clinical trial. Malignant melanomas and adenocystic carcinomas of head and neck well respond to the HIMAC carbon ion radiotherapy, while squamous cell carcinomas of head and neck show a moderate response to the carbon ions. This has implied that malignant melanomas in general could have larger RBE to carbon ions than squamous cell carcinomas. We collected more than 10 cell lines each for human malignant melanomas and squamous cell carcinomas, and compared dose responses between carbon ions and X rays. Growth of malignant melanomas and adenocystic carcinomas is slower than squamous cell carcinomas. As RBE of fast neutrons depends on the doubling time of lung tumors in human, the second question should also be answered by using different tumors with different growth rates. We used 10 transplantable tumors in C3H mice, and compared RBE values of carbon ions.
