内容記述 Evidence has accumulated demonstrating bystander effects where ionization events in one cell can be sensed in neighbouring cells. Most of the previous work used either attached cells or three-dimensional cultures as models to study radiation-induced bystander responses, and little literature can be found for suspension cells. In the present study, unirradiated L5178Y (LY) suspension cells were co-cultured for 16 h with neoplastic human salivary gland (HSG) cells which had been irradiated either with 290 MeV/u carbon ions or X-rays. The results showed that the survival of the unirradiated recipient LY cells was biphasic and depended on the dose delivered to the co-cultured HSG cells. Cell damage measured as apoptosis and necrosis was also detected in a 48 h subculture of the unirradiated LY cells and its induction increased but then had a tendency of decrease when the irradiation dose delivered to the HSG cells increased. When PTIO, a nitric oxide (NO)-specific scavenger, was present in the co-culture medium, the cellular damage in the unirradiated LY cells was obviously diminished to a low level, suggesting that NO contributes to this bystander damage. This idea was strengthened by the finding that, an oxidization product of NO, nitrite was produced in the co-culture medium. The time course of nitrite induction corresponded to that of the viability of irradiated HSG cells, which indicated that NO was secreted from the irradiated HSG cells during their programmed death. Irradiation of HSG cells with high LET was found to be much more effective in producing nitrite and further inducing damage to the unirradiated recipient LY cells than low LET irradiation. Moreover, the relationship of the concentration of nitrite in the co-culture medium to the survival and the cell damage of the unirradiated LY cells in turn pointed to the involvement of NO in the above-mentioned bystander responses. This mechanistic study may have significance for radiotherapy and low-level environmental radiation.
