In this study, we examined the possibility of radioiodinated anti Tenascin-C antibody Fab\u0027 fragment (125I-TN-C) for estimating therapeutic efficacy of carbon ion therapy (270 Mev, 25 Gy) to C-10 glioma xenografts in nude mice. In addition, since radioiodinated 3-[125I]iodo-alpha-methyl-L-tyrosine (125I-IMT) is currently being used as a diagnostic regent of brain tumor, therapeutic effects of ion therapy was also estimated using the radiopharmaceutical. After carbon beam irradiation of left leg of nude mice without tumor, the damage of irradiation site was increased with times. The uptake of 125I-TN-C in irradiation site was also increased (irradiation site-to-non-irradiation site 4.15+/-0.95). At 10 and 20 days post-carbon ion therapy, however, no significant differences were observed between the tumor site and control leg. The radioactivity levels in the tumor were lower than those in the blood. On the other hand, 125I-IMT accumulated in tumor but not in irradiation site. The tumor-to-blood and the irradiation site-to-non-irradiation site ratios of the radioactivity were 5.96+/-1.00 and 1.57+/-0.70, respectively.
