内容記述 To determine radiobiological mechanisms for high-LET radiation and repair kinetics of DNA double-strand breaks (dsb) as a function of LET and/or ion-species, V79 and DNA-dsb repair gene knokuout DT40 (chicken leukemia B cells; ku70-/-, rad54-/-, ku70-/-rad54-/-, and the parent DT40) cells were exposed to heavy-ion beams at HIMAC. The LET dependences of cell killing and chromosome aberrations on the knockout cells were determind. Also SLDR from heavy-ion induced damage in V79 cells were investigated using X-rays as secondary radiation. LET dependencies of production of chromo-some fragments in DT40 cells showed a big difference revealed by the difference in presence or absence of the repair genes in the cell. An analogous LET-RBE relationship to cells killing was observed. There is any peak of RBE at around 100-200 keV/um for the repair deficient cells. This means that increase of the sensiticity can be seen only for the repair suficient cells, and suggests that the peak of the RBE caused by misrepair of DNA dsb. LET dependency of SLDR or change in the shoulder of survival curves on V79 cells were determined by secondary X-ray beams following exposures of different high-LET beams as primary irradiation and different repair time. No change in the shoulder for secondary X-rays was found among all (up to 800 keV/um) primary high-LET irradiations. This indicates SLD produced by heavy-ions are completely repaired by the repair system.
