| 内容記述 | The purpose of this study is to investigate the normal tissue damage caused by X-ray or carbon-ion therapy. We measured chromosomal aberrations in peripheral blood lymphocytes before, during and after the radiation treatment by using the novel technique of Interphase Chromosome Painting. Premature chromosome condensation(PCC) was induced in stimulated lymphocytes by incubation in calyculin A and sledes were hybridized in situ with whole-chromosome 2 and 4 (FISH). Chromosome aberrations decreased after the end of radiiotherapy. The fraction of complex-type chromosomal exchanges presented a good correlation with the acute toxicity in esophageal cancer patients. The initial slope of the dose response curve for the induction of chromosomal aberrations during the treatment was correlated to the relative decrease in the number of white blood cells and lymphocytes during the treatment. In lung cancer patients treated with carbon-ion, the frequency of was correlated to the number of lymphocytes and white blood cells following radiation therapy. These results suggest that Interphase Chromosome Painting (PCC+FISH) is an efficient tool for biodosimetry of radiation treatmen. |
